大豆GH3家族三个成员对20种农药的转录响应

摘 要:

植物GH3基因家族具有生长素(IAA)等植物激素的氨基酸化合成酶活性, 参与调节IAA等的动态平衡, 在植物生长发育和抗逆性方面具有重要作用。为了明确大豆GH3基因家族成员对主要农药的响应特点, 用qRT-PCR分析了20种不同作用机制的农药对该家族XM003518758XM003521754XM003543231在60 min内的诱导特点。结果表明, XM003518758基因的表达受草甘膦、氰戊菊酯轻微抑制, 受2甲4氯等其余18种农药诱导上调但不超过2.5倍。所有测试的20种农药均不抑制XM003521754基因的表达, 而最大诱导表达量达10倍以上。2甲4氯、乙草胺、百草枯、氰戊菊酯和戊唑醇对XM003543231诱导表达超过50倍, 而其余15种测试农药对XM003543231的表达无抑制效应。而大豆XM003521754基因只受2甲4氯、二氯喹啉酸和多菌灵的显著诱导上调。

Abstract:

GH3 gene family plays prominent roles in plant development and stress resistance through maintaining phytohormone homeostasis depended on their IAA-amido synthetase activity. In order to explore the expression patterns in transcription level of the soybean GH3 gene family, we used qRT-PCR to determine the response of the three members of the family, XM003518758, XM003521754 and XM003543231, after exposure to 20 pesticides within 60 minutes. The expression of XM003518758 was slightly depressed by glyphosate and cyhalothrin, and induced no more than 2.5 times by the other 18 pesticides tested. All 20 pesticides tested were not depressed the expression of XM003521754 and some of them induced the XM003521754 expression over 10 times. Over 50 times up-regulated expression of XM003543231 was induced by MCPA, acetochlor, gramoxone, cyhalothrin, and tebuconazole, while the other 15 pesticides tested had not depressed-effect on the expression of XM003543231. It was important that the gene XM003521754 was induced only by auxin-like pesticides as MCPA, quinclorac, carbendazim.
